Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (low-affinity Fc gamma RIII). CD16 exists as two isoforms, FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, NK cells.
• FcγRIIIB: Expressed on neutrophils (GPI-anchored form).

Binding of IgG triggers signal transduction pathways leading to ADCC, phagocytosis, cytokine release, and antigen presentation.
3G8 recognizes the same epitope as CB16 but distinct from B73.1.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells.
Use at 4 µL (0.6 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Staining Buffer Recommendation:
Each NovaFluor conjugate ships with CellBlox Blocking Buffer.

• Add 5 µL per sample when staining with NovaFluor conjugates.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

CD16 encodes the Fc receptor for IgG involved in immune complex clearance, ADCC, and viral infection enhancement.

• Isoforms: FCGR3A (NK cells, transmembrane anchored), FCGR3B (neutrophils, PI-anchored)
• Associated diseases: Immunodeficiency 20, Herpes Zoster, systemic lupus erythematosus, alloimmune neonatal neutropenia.
  Alternative splicing generates multiple isoforms.
