Target Information
Pyridoxal kinase (PDXK) converts vitamin B6 to pyridoxal-5-phosphate (PLP), an essential cofactor in amino acid and neurotransmitter metabolism. The PDXK gene encodes a 312-amino acid polypeptide, and its expression demonstrates pyridoxal kinase activity. A major 1.5-kb PDXK transcript is expressed in all tissues tested, with circadian oscillations in expression. In mice, liver and brain Pdxk expression is regulated by the PAR bZIP transcription factors Dbp, Hlf, and Tef, which also show circadian oscillations. Mice lacking these transcription factors exhibit reduced brain PLP, serotonin, and dopamine levels, resulting in susceptibility to spontaneous and audiogenic epilepsies.
