Target Information

RARA is a receptor for retinoic acid. Retinoic acid receptors bind as heterodimers to their target response elements in response to ligands such as all-trans or 9-cis retinoic acid, regulating gene expression in various biological processes. RXR/RAR heterodimers bind to retinoic acid response elements (RARE) composed of tandem 5'-AGGTCA-3' sites (DR1–DR5).
In the absence of ligand, RXR-RAR heterodimers associate with transcription corepressor complexes that induce histone acetylation, chromatin condensation, and transcriptional suppression. Upon ligand binding, corepressors dissociate and coactivators are recruited, leading to transcriptional activation.
RARA plays an essential role in retinoic acid-induced germ cell development during spermatogenesis and supports early spermatocyte survival. In Sertoli cells, it promotes development of early meiotic spermatocytes. Together with RARG, it is required for skeletal growth, matrix homeostasis, and growth plate function. It regulates target gene expression via ligand-dependent recruitment of chromatin complexes containing KMT2E/MLL5 and mediates retinoic acid-induced granulopoiesis.
