Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a transcription factor crucial for thymic development and Th2 differentiation.
Expression during embryonic development occurs in the CNS, skin, mammary glands, and kidney. Homozygous knockout of Gata-3 is embryonic lethal.
In the thymus, Gata-3 is mainly expressed in CD4 single positive cells and regulates Th2 differentiation by binding to the IL-4 promoter and repressing T-bet expression, thus inhibiting Th1 differentiation.

Alternative splice variants have been reported, especially in the MCF7 cell line. The TWAJ antibody recognizes both 50 and 45 kDa forms.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Cross-reactivity in rhesus monkeys has been documented.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometry using mouse thymocytes with the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523).

Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Target Information

GATA3 is a zinc finger transcription factor controlling T-cell receptor (TCR) gene activation. It binds enhancer elements of all four TCR genes (alpha, beta, gamma, delta).
Expressed in T cells, not in B cells or macrophages, GATA3 is essential for T-lymphocyte lineage development.
The human GATA3 gene comprises six exons over 17 kb, encoding two zinc fingers conserved with GATA1.
