Target Information

LATS2 is a serine/threonine protein kinase belonging to the LATS tumor suppressor family. It interacts with a negative regulator of p53 and functions in a positive feedback loop with p53 in response to cytoskeleton damage, leading to centrosome and mitotic apparatus regulation. LATS2 plays an essential role in maintaining mitotic fidelity and genomic integrity, and is pivotal in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The LATS signaling pathway includes a kinase cascade involving STK3/MST2 and STK4/MST1 with SAV1, which activate LATS1/2 with MOB1, leading to phosphorylation and inactivation of YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 prevents its nuclear translocation, regulating genes related to cell proliferation, death, and migration. It also negatively regulates the G1/S transition by down-regulating cyclin E/CDK2 kinase activity.
