Cullin 4B (CUL4B)는 다양한 cullin-RING 기반 E3 유비퀴틴-단백질 리가제 복합체의 핵심 구성 요소로서, 표적 단백질의 유비퀴틴화 및 프로테아좀 분해를 매개합니다. 복합체의 기능적 특이성은 기질 인식 서브유닛에 따라 달라집니다.
CUL4B는 복합체 내에서 scaffold 단백질로 작용하여 기질과 유비퀴틴 결합 효소의 위치를 조정함으로써 촉매 작용을 돕습니다. 방사선 유도 DNA 손상 시 CDT1, 히스톤 H2A, H3, H4의 다유비퀴틴화를 매개하며, DDB2에 의해 UV 손상된 크로마틴으로 이동하여 DNA 복구 및 복제에 중요합니다. 또한 cyclin E의 유비퀴틴화를 통해 정상적인 G1 세포주기 진행을 조절하고, mTOR 경로를 통해 세포 성장과 대사 조절에 관여합니다.
CUL4B의 mTORC1 매개 경로 조절은 26S 프로테아좀 기능에 의존하며 MLST8과의 상호작용을 필요로 합니다.
