Target Information

CRTH2 (GPR44) is a chemoattractant receptor-homologous molecule expressed on Th2 cells that acts as a receptor for prostaglandin D2 (PGD2) and prostaglandin D receptor. It mediates chemotaxis of Th2 cells and eosinophils to PGD2, playing a crucial role in allergic responses.

CRTH2 knockout mice showed enhanced eosinophil recruitment and increased IL-5 and IL-3 production, suggesting CRTH2 restricts eosinophilia and allergic responses in vivo. It is a member of the G protein-coupled leukocyte chemoattractant receptor family, selectively expressed in Th2 lineage cells.

CRTH2 expression has been reported in the central nervous system, peripheral tissues, and blood cells, particularly in activated Th2 cells. Two transcripts have been identified:

• 3.5 kb transcript: thalamus, frontal cortex, pons, hippocampus
• 3.4 kb transcript: fetal liver, leukocytes, thymus

CRTH2 is expressed in activated Th2 cells, including allergen-responsive Th2 cells, indicating its pivotal role in Th2-type immune reactions. At least two isoforms of CRTH2 are known. SNPs in the 3′ UTR of the CRTH2 gene have been associated with asthma susceptibility.
