This product is diluted and ready to use.
A recommended positive control tissue is Uterus, though other tissues may also be suitable.
Designed for professional laboratory use in detecting Caldesmon in formalin-fixed, paraffin-embedded tissue via manual qualitative IHC testing.
Intended for use after initial histopathologic tumor diagnosis.
Caldesmon is a developmentally regulated protein involved in smooth and non-muscle contraction. Two variants exist:
- h-Caldesmon (120–150 kDa): Predominantly in smooth muscle
- l-Caldesmon (70–80 kDa): Found in non-muscle tissue and cells
Neither variant is detected in skeletal muscle.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential antibody applications, enzyme complexes, and chromogenic substrates.
Results are interpreted microscopically to aid in differential diagnosis of antigen-associated pathophysiological processes.
A positive tissue control must accompany each staining procedure.
- Should include both positive and negative staining components
- External positive controls: freshly processed autopsy/biopsy/surgical specimens
- Weak positive staining preferred for detecting subtle changes in antibody sensitivity and reagent stability
Negative control tissues (internal or external) ensure absence of non-specific staining.
If specific staining occurs in negative controls, results from patient specimens are invalid.
