Product Specific Information

Description:
This 12.14 monoclonal antibody reacts with mouse Nur77 (NR4A1, TR3, NGFI-B, NAK1), an inducible orphan nuclear receptor expressed in thymocytes and T cell lines. Nur77 promotes apoptosis and is involved in thymocyte negative selection. It also plays roles in steroid biosynthesis in Leydig cells, dopamine response, and interaction with FoxP3 in regulatory T cells.

Applications Reported:
For intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometry of stimulated mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.
Refer to BestProtocols®: Protocol B for intracellular (nuclear) proteins.
Recommended ≤1 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Optimize concentration for best results.

Target Information

NR4A1 (Nur77):
A member of the steroid-thyroid hormone-retinoid receptor superfamily. Expressed in brain, adrenal, thyroid, liver, testis, ovary, thymus, muscle, lung, and prostate. Induced by stress stimuli and growth factors. Functions as a nuclear transcription factor mediating apoptotic signaling and hypothalamic-pituitary axis signaling. Translocation from nucleus to mitochondria induces apoptosis. Multiple transcript variants encode distinct isoforms; RXR is a known partner for NR4A1.
