Product Specific Information

This 4H9 monoclonal antibody reacts with human CD7 (Leu-9, gp40), a 40-kDa transmembrane glycoprotein belonging to the immunoglobulin superfamily. CD7 is expressed on peripheral blood T lymphocytes and natural killer cells, as well as on lymphoid and myeloid lineage precursors during adult hematopoiesis. It is involved in T cell activation through association with CD3 and CD45.

CD7 is also detected in T cell-derived leukemias and lymphomas. Because CD7 expression is greater on CD4+ T cells in classical Hodgkin’s lymphoma than in benign reactive lymphadenopathy, it may serve as a diagnostic marker for Hodgkin’s lymphoma.

Blocking assays demonstrate that this clone recognizes the same epitope as clone 124-1D1.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.125 µg) per test. A test is defined as the amount of antibody staining a sample in a final volume of 100 µL.

Target Information

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily. The N-terminal region (amino acids 1–107) shows high homology to Ig kappa-L chains, while the C-terminal region is proline-rich and forms a stalk projecting the Ig domain. CD7 is expressed on most immature and mature T lymphocytes, T cell leukemia cells, NK cells, and a small subset of B cells. It positively modulates T cell and NK cell activity through calcium fluxes, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates directly with phosphoinositol 3′-kinase, and its ligation induces D-3 phosphoinositide production and tyrosine phosphorylation. CD7 expression serves as an important marker in leukemia diagnostics.
