Target Information
Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering.
The system’s flexibility arises from its RNA-dependent targeting mechanism, enabling precise genome editing such as indel mutations, sequence insertions, deletions, and rearrangements.
Cas9 variants include Streptococcus pyogenes (SpCas9) and Staphylococcus aureus (SaCas9). SaCas9 offers advantages such as smaller size, high efficiency, nickase activity, and specificity.
SaCas9 recognizes an NNGRRT PAM sequence and cleaves target DNA with spacer lengths of 20–24 nucleotides, comparable to SpCas9. Due to its compact size (1053 AA vs. 1368 AA for SpCas9), SaCas9 is preferred for all-in-one delivery systems using adeno-associated vectors.
