A recommended positive control tissue is rejected kidney, though other tissues may also serve as positive controls.
The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue stained by manual qualitative IHC testing. It is used after the primary diagnosis of tumors made by conventional histopathology.
Complement component proteins (C2, C3, C4, C5) are potent anaphylatoxins released during complement activation. Binding to their respective receptors (C3aR, C1R, C5aR) triggers proinflammatory responses such as cellular degranulation, smooth muscle contraction, cytokine release, and leukocyte activation.
C3d, a terminal degradation product of C3, modulates adaptive immune responses through interaction with complement receptor type 2 (CR2), linking innate and adaptive immunity.
For optimal staining, pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. IHC staining visualizes antigens via sequential application of primary and secondary antibodies, enzyme complexes, and chromogenic substrates, followed by washing steps. The reaction product is observed under a light microscope to aid in differential diagnosis.
A positive tissue control must accompany each staining procedure. This ensures proper tissue preparation and staining quality. External positive controls should be freshly prepared and processed similarly to patient samples. Weakly positive tissues are recommended for quality assurance to detect minor reagent degradation.
Negative controls (internal or external) should confirm absence of specific staining and help assess background noise. If specific staining appears in negative controls, patient results are invalid.

C3 is a key component of the complement system where classical and alternative activation pathways converge at the C3 activation step. C3 is cleaved into C3a and C3b.
The molecular mass of C3 is 185 kDa, composed of two chains (110 kDa and 75 kDa) linked by disulfide bonds.
