Target Information
The LRP2 (low density lipoprotein-related protein 2), also known as megalin, is a multi-ligand endocytic receptor primarily expressed in absorptive epithelial tissues such as the kidney.
It features a large extracellular domain, a single transmembrane domain, and a short cytoplasmic tail.
LRP2 binds various macromolecules including albumin, apolipoproteins B/E, and lipoprotein lipase, and is essential for reuptake of lipoproteins, sterols, vitamin-binding proteins, and hormones.
It also participates in cell signaling, interacting with ligands such as parathyroid hormones and sonic hedgehog, and cytosolic partners like MAP kinase scaffold proteins.
Receptor recycling is regulated via phosphorylation of the cytoplasmic domain.
Mutations in this gene are associated with Donnai-Barrow syndrome (DBS) and facio-oculoacoustico-renal syndrome (FOAR).
