• Reconstitution: Reconstitute with 0.5 mL of distilled water.
• Dilution: Use a buffer containing carrier protein (e.g., PBS with 1% BSA) or a neutral medium containing 10% FCS. Store aliquots at -70°C.
• Activity: The biological activity of recombinant canine TNF-alpha was measured by its cytolytic effect on mouse WEHI-13VAR cells in the presence of Actinomycin D. The ED50 (or 1 unit of activity) is typically 10 pg/mL.

Target Information

TNF-alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.

It exists as multimers (dimers, trimers, or pentamers) but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. TNF-alpha demonstrates tumor necrosis activity in vivo and plays a key role in immune responses to bacterial, viral, parasitic, and fungal infections. It is synergistic with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
