This antibody is supplied in a ready-to-use formulation.
Recommended positive control tissue: Breast adenocarcinoma (other tissues may also be suitable).

The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue stained via manual qualitative IHC.
It is used after tumor diagnosis by conventional histopathology.

Recognizes a 67 kDa estrogen receptor (ER) alpha protein. The ER gene spans over 140 kb, divided into 8 exons, and encodes a protein with six domains (A–F). This antibody strongly stains the nucleus of epithelial cells in breast carcinomas.
ER presence in breast tumors indicates a higher likelihood of response to antiestrogen therapies (e.g., tamoxifen).

Recommended Use

• Use with formalin-fixed, paraffin-embedded sections.
• Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
• Positive tissue controls must be included in every staining procedure.
• Weakly positive tissues are preferred for external positive controls to ensure sensitivity and detect reagent degradation.
• Internal or external negative controls should confirm absence of non-specific staining.

Target Information

Estrogen Receptors (ER) are nuclear receptors in the steroid/thyroid hormone receptor family.
Upon estrogen binding, ER undergoes conformational change, dimerizes, and binds to Estrogen Response Elements (EREs) in DNA, regulating transcription through coactivators or corepressors.

Two main forms exist:

• ERα (ESR1)
• ERβ (ESR2)

Alternative splicing produces at least four ERα isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa).
ERs are widely expressed and play key roles in sexual development, reproductive function, bone health, and are implicated in breast cancer, endometrial cancer, and osteoporosis.
