Target Information
FBXW7 (F-Box and WD Repeat Domain Containing 7) is a member of the F-box protein family, characterized by a ~40 amino acid F-box motif. These proteins form part of the SCF (SKP1–Cullin–F-box) ubiquitin ligase complex, which mediates phosphorylation-dependent ubiquitination.
The F-box proteins are categorized into:
- Fbws: Containing WD-40 domains
- Fbls: Containing leucine-rich repeats
- Fbxs: Containing other interaction modules or no recognizable motifs
FBXW7 (also known as FBX30) belongs to the Fbws class and includes an F-box and seven tandem WD40 repeats. It binds directly to cyclin E, targeting it for ubiquitin-mediated degradation. Mutations in FBXW7 have been identified in ovarian and breast cancer cell lines, suggesting a role in tumorigenesis. Multiple transcript variants encoding different isoforms exist.
