Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포사멸, 분화 등 다양한 세포 과정을 조절합니다. SMAD2는 TGF-beta 수용체와 상호작용하며, 핵으로의 이동은 TGF-beta 신호전달의 핵심 단계입니다.
ERK1에 의해 MH1 도메인의 칼모듈린 결합 영역 내 Thr8가 인산화되면 SMAD2의 전사 활성은 증가하며, 이는 칼모듈린에 의해 음성 조절됩니다.
TGF-beta 신호에 반응하여 SMAD2는 수용체에 의해 인산화되고 SARA와의 결합이 해리되며 SMAD4와 결합합니다. SMAD4와의 결합은 핵으로의 이동과 표적 프로모터 결합, 전사 억제 복합체 형성을 위해 중요합니다.
또한 SMAD2는 activin type 1 receptor kinase에 의해 인산화될 수 있으며 activin 신호를 매개합니다.
