Juvenile nephronophthisis type 1은 NPHP1 유전자 돌연변이에 의해 발생하며, 이 유전자는 nephrocystin을 암호화합니다.
Nephrocystin은 p130Cas (BCAR1), PTK2B (Pyk2), tensin과 상호작용하며, 배아 신장과 고환에서 공통 신호 전달 경로에 관여합니다.
Nephrocystin과 p130Cas는 세포 간 접촉 부위 근처에서 함께 발현되며, nephrocystin의 발현은 Pyk2의 Tyrosine 402 인산화를 유도하고 ERK1/ERK2 같은 MAP kinase를 활성화합니다.
Nephrocystin은 진화적으로 보존된 SH3 도메인을 포함합니다.
