Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein. CD21 is expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells. It serves as a receptor for complement component C3d and Epstein-Barr virus (EBV). CD21, along with CD19 and CD81, forms a multimolecular complex with BCR involved in B cell activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.05 µg) per test (100 µL total volume, 10⁵–10⁸ cells).

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor) binds C3 complement fragments and plays roles in immunocomplex uptake, memory B cell survival, and humoral immune response development. It also serves as a receptor for EBV, contributes to prion targeting, and forms part of a signal-transduction complex with CD19, CD81, and Leu13 for B cell activation. Genetic variations in CD21 are associated with systemic lupus erythematosus type 9 (SLEB9). Alternative splicing yields multiple isoforms.
