Product Specific Information

Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C followed by cooling at room temperature for 20 minutes.

Target Information

CD63 (LAMP-3, lysosome-associated membrane protein-3) is a glycoprotein of the tetraspanin family, found in late endosomes, lysosomes, and secretory vesicles. It is also present on the plasma membrane following cell activation and is widely used as a basophil activation marker.
In mast cells, CD63 exposure does not require activation. It interacts with integrins, influences phagocytosis and cell migration, and is involved in H/K-ATPase trafficking and regulation of ROMK1 channels. CD63 also acts as a T-cell costimulation molecule and can be used as a prognostic marker in early-stage carcinomas.
CD63 is expressed on activated platelets and translocated to the plasma membrane after activation. It is also found in monocytes, macrophages, and weakly expressed on granulocytes, B, and T cells. CD63 is identical to melanoma-associated antigen ME491 and platelet antigen PTLGP40.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
