Target Information
MARCH8 (c-MIR) is a novel E3 ubiquitin ligase belonging to the modulator of immune recognition (MIR) family, characterized by a variant RING-CH domain. It functions as a homolog of KSHV MIR1 and MIR2, targeting B7-2 for lysosomal degradation and down-regulating its surface expression through ubiquitination. MARCH8 also down-regulates transferrin receptor and Fas, impacting apoptosis induction. It is the first E3 ubiquitin ligase known to inhibit MHC II expression. Recent studies suggest MARCH8 regulates immune responses by promoting ubiquitination of MHC-II and CD86, leading to endocytosis and lysosomal degradation.
