Target Information
ZRANB3 is a DNA annealing helicase and endonuclease required to maintain genome stability at stalled or collapsed replication forks by facilitating fork restart and limiting inappropriate recombination during template switching events.
It is recruited to stalled DNA replication sites by polyubiquitinated PCNA and acts as a structure-specific endonuclease that cleaves replication fork D-loop intermediates, generating an accessible 3'-OH group for DNA polymerase extension.
In addition to endonuclease activity, ZRANB3 catalyzes fork regression via annealing helicase activity to prevent fork disintegration and double-strand break formation.
