Target Information

Signal transduction of TGF-beta superfamily members is regulated by Smad proteins. Smads relay signals from the cell membrane to the nucleus, influencing specific gene transcription. Smad4 plays an essential role in TGF-beta-induced transcriptional activation, where phosphorylated receptor-associated Smads associate with Smad4.
SMIF (Smad4-interacting protein) and Smad4 form complexes with TGF-beta and BMP4, facilitating nuclear translocation. The SMIF-Smad4 interaction occurs via an EVH1/WH1 domain on SMIF and a proline-rich activation domain on Smad4. Deletion of the Smad4-interacting domain in SMIF abolishes TGF-beta-induced nuclear translocation. The human SMIF gene encodes a 70 kDa protein expressed ubiquitously.
