Background
Aminoacyl-tRNA synthetases are enzymes that charge tRNAs with their cognate amino acids. The HARS gene encodes a cytoplasmic class II aminoacyl-tRNA synthetase responsible for histidyl-tRNA synthesis, essential for histidine incorporation into proteins. It is located in a head-to-head orientation with HARSL on chromosome 5, sharing a bidirectional promoter. The gene product is a frequent autoantibody target in polymyositis/dermatomyositis, and multiple transcript variants exist.
