Background
This gene encodes the ghrelin-obestatin preproprotein that is cleaved to yield two peptides, ghrelin and obestatin. Ghrelin is a powerful appetite stimulant and plays an important role in energy homeostasis. Its secretion begins when the stomach is empty, binding to the growth hormone secretagogue receptor in the hypothalamus, resulting in growth hormone (somatotropin) secretion. Ghrelin is involved in hunger regulation, reward perception, gastric acid secretion, gastrointestinal motility, and insulin secretion. Obestatin, derived from the same precursor, was initially thought to counteract ghrelin by promoting satiety, though its exact role remains under debate.
