Target Information
MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily.
It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway and regulates T cell receptor (TCR) signaling and proliferation by phosphorylating the adaptor protein SLP-76.
HPK1 plays a crucial role in stress responses and blood cell differentiation. It is predominantly expressed in hematopoietic cells, including early progenitor cells, and is related to p21(Cdc42/Rac1)-activated kinase (PAK) and yeast STE20.
HPK1 specifically activates JNK1 and enhances AP-1-mediated transcriptional activity in vivo. Unlike PAK65, HPK1 does not bind to Rac1 or Cdc42.
