Target Information
IRS-1 (Insulin Receptor Substrate 1) is a major substrate of the insulin receptor. It is phosphorylated in response to insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4) stimulation.
IRS-1 undergoes phosphorylation on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates a portion of insulin-stimulated serine/threonine phosphorylation in vivo, with Thr502 and Ser99 identified as key phosphorylation sites.
IRS-1 contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with the Ser270 motif located in the phosphotyrosine-binding domain responsible for interaction with the insulin receptor.
