Target Information
STK17B (also known as DRAK2) is a member of the serine/threonine kinase family related to death-associated protein kinase that triggers apoptosis. It is selectively important for T-cell survival, and inhibition of STK17B has therapeutic potential for autoimmune diseases. T-cell survival depends on balanced receptor and co-stimulatory signals; deficiency of STK17B can affect autoimmune susceptibility without general immune suppression.
Protein kinases are key in signal transduction responding to external stimuli. Several kinases, including ZIP kinase, DRAK1, and DRAK2, are associated with apoptosis. Overexpression of these kinases in NIH 3T3 cells induces apoptotic morphological changes. The kinase domains of DRAKs and ZIP kinase are homologous to DAP kinase, which is involved in interferon-γ–induced apoptotic signaling. These findings suggest that the C-terminal domain of DRAKs is crucial for apoptosis induction.
