Target Information
Epidermal Growth Factor (EGF) binds to the EGF receptor (EGFR), a 170 kDa protein kinase, initiating diverse cellular pathways. Upon activation, EGFR forms homo- or heterodimers, each triggering distinct signaling cascades through recruitment of SH2-containing effector proteins. EGF is widely expressed in kidney, brain, prostate, and salivary glands, acting as a potent mitogenic factor. The phosphorylated receptor recruits adapter proteins such as GRB2, activating downstream signaling cascades including RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, and STAT pathways. EGF may also influence NF-κB signaling. Dysregulation of the EGF gene is associated with hypomagnesemia type 4 and certain cancers.
