Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a protein that localizes with tight junction proteins such as ZO-1 and occludin in polarized epithelial cells. It is not a structural component but is recruited during tight junction maturation. Metadherin is overexpressed in breast cancer tissues and xenografts, while expression in normal breast tissue is low. It binds to lung vasculature—one of the common sites of breast cancer metastasis—via a C-terminal extracellular domain segment. Blocking this domain with antibodies or inhibiting MTDH via siRNA has been reported to reduce breast cancer metastasis.
