Target Information
KLOTHO is a systemic anti-aging hormone belonging to the glycosidase1 superfamily. It encodes a type I membrane protein abundant in the kidney and brain. In mice, KLOTHO deficiency leads to phenotypes resembling human aging, including arteriosclerosis, osteoporosis, skin atrophy, growth retardation, short lifespan, and infertility. Overexpression of KLOTHO extends lifespan by inhibiting insulin/IGF1 signaling. KLOTHO also regulates calcium and phosphorus homeostasis by inhibiting active vitamin D synthesis and acts as a potential tumor suppressor.
