Target Information
The protein encoded by the PTRH2 gene is a mitochondrial protein with two putative domains: an N-terminal mitochondrial localization sequence and a UPF0099 domain. In vitro assays suggest that this protein exhibits peptidyl-tRNA hydrolase activity, releasing the peptidyl moiety from tRNA to prevent accumulation of dissociated peptidyl-tRNA, thereby maintaining translation efficiency. PTRH2 also regulates cell survival and death, promoting survival as part of an integrin-signaling pathway in cells attached to the extracellular matrix. Mutations in this gene are associated with intellectual disability, postnatal microcephaly, progressive cerebellar atrophy, hearing impairment, polyneuropathy, failure to thrive, and organ fibrosis with exocrine pancreas insufficiency. Alternative splicing results in multiple transcript variants encoding different isoforms.
