Target Information
The CDKN2A gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which act as inhibitors of CDK4 kinase. Another transcript encodes an alternate open reading frame (ARF) that stabilizes the tumor suppressor protein p53 by interacting with and sequestering MDM2, an E3 ubiquitin-protein ligase responsible for p53 degradation. Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product share a role in cell cycle G1 control through regulation of CDK4 and p53. This gene is frequently mutated or deleted in various tumors and is recognized as an important tumor suppressor gene.
