Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing key roles in cell death regulation and stress response.
TRAF proteins share a conserved region enabling receptor and protein interactions, activating distinct signaling cascades.
TRAF2 and TRAF3 bind to CD40, crucial for NF-κB activation in B lymphocytes, while TRAF2 and TRAF6 are essential for CD40 signaling in non-hematopoietic cells.
Additionally, TRAF2 interacts with lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
