Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was originally identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates various cellular responses such as proliferation, activation, differentiation, and apoptosis.

CD30 plays a key role in the pathophysiology of Hodgkin’s disease and other CD30+ lymphomas, functioning as a costimulatory molecule in thymic negative selection. It is expressed on Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (including Burkitt’s), virus-infected T and B cells, and on activated normal T and B cells.

In T cells, CD30 is found on subsets that produce Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. The soluble form (sCD30) serves as a marker for Th2 immune response. CD30 interacts with TRAF2 and TRAF5, mediating NF-κB activation and promoting apoptosis. It also limits autoreactive CD8 effector T cell proliferation, contributing to immune regulation. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.

CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL). It is also expressed by embryonal carcinomas.
