Target Information
Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
This enzyme plays a crucial role in folate homeostasis in mammals by:
- Preventing efflux of folate cofactors from the cell via polyglutamation.
- Increasing binding of folate cofactors to enzymes involved in folate interconversion and biosynthesis.
- Allowing accumulation of folates in mitochondria necessary for glycine synthesis.
FPGS is widely expressed in human tumors and is associated with cellular proliferation or lack of differentiation. Its activity contributes to the efficiency of folate antimetabolites used in cytotoxic therapies.
