Target Information
The pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to execute genetic programs. Several pax genes are associated with developmental mutations, such as:
- pax-3: linked to Waardenburg syndrome
- pax-6: linked to Aniridia
- pax-2: linked to Wilms tumor
During embryogenesis, Pax-2 is expressed in the developing kidney, specifically in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Pax-2 expression decreases as the tubular epithelium matures but persists in the undifferentiated epithelium of Wilms tumors, correlating with proliferation of poorly differentiated epithelial cells. Expression of the Wilms tumor suppressor protein WT1 coincides with down-regulation of pax-2, suggesting WT1 directly represses pax-2 transcription.
