Target Information

BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family can form dimers to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak are critical for cytochrome c release from mitochondria, initiating apoptosis.
Constitutive expression of BCL-2, such as through translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas normal germinal centers do not. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 has been linked to cancers including B-cell lymphoma and prostate cancer.
