Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is crucial for p53 activation and stabilization. p53 functions as an anticancer agent by preventing proliferation of damaged cells. Phosphorylation at Ser392 has been associated with tumor formation. Elevated p53 levels are linked to aging, oxidative stress, and memory deficits. While normal cells express low levels of p53, transformed cell lines show high expression contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor activity, occurring both somatically and germline (e.g., Li-Fraumeni syndrome).
