Target Information
The NF-κB/Rel transcription factors are present in the cytosol in an inactive state, complexed with inhibitory IκB proteins. Activation of NF-κB typically occurs through phosphorylation-induced, proteasome-mediated degradation of IκB. This process involves activation of a high molecular weight IκB kinase (IKK) complex composed of three subunits: IKKα, IKKβ, and IKKγ. IKKα and IKKβ serve as catalytic subunits, while IKKγ acts as a regulatory subunit. Activation of IKK requires phosphorylation at Ser177 and Ser181 in IKKβ (Ser176 and Ser180 in IKKα), leading to conformational changes and kinase activation.
