Target Information

Neurofilaments (NFs) are a type of intermediate filament (IF) expressed almost exclusively in neuronal cells, most prominently in large axons. NFs in vertebrates are composed of three different polypeptide chains — neurofilament medium protein (NF-M), high (NF-H), and light protein (NF-L). These share sequence and structural similarity in a coiled-coil core domain but differ in the length and sequence of their termini.

NF-M protein tail-mediated interactions are critical for axonal size and cytoskeletal architecture, mediated by the highly phosphorylated tail domain. NF-M phosphorylation and O-GlcNAcylation are reciprocally regulated, affecting translocation, filament formation, and function. Antibodies to neurofilament subunits are useful neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
