Target Information
Biological actions of insulin and IGF-1 are mediated by their respective cell surface receptor tyrosine kinases that regulate multiple signaling pathways through activation of phosphorylation cascades. Insulin/IGF-1 binding to the extracellular domain leads to autophosphorylation of downstream target proteins. These receptors differ in sequence regions conferring ligand specificity and intracellular signaling domains, resulting in distinct functional outcomes. Defects in IR are associated with insulin resistance syndromes, while IGF-1R defects may lead to growth retardation. Both signaling cascades are important in cancer development.
