Target Information
The human AF9 (MLLT3) gene is a frequent fusion partner with the ALL1 (MLL) gene at 11q23, resulting in the t(9;11)(p22;q23) translocation. The AF9 gene spans over 100 kb and contains two patient breakpoint cluster regions (BCRs):
- BCR1 in intron 4 (site A)
- BCR2 spanning introns 7 and 8 (site B)
AF9 includes several structural elements such as:
- A colocalizing in vivo DNA topoisomerase II cleavage site
- An in vitro DNase I hypersensitive site in intron 7 (BCR2)
- Two scaffold-associated regions (SARs): SAR1 (intron 4) and SAR2 (exons 5–7)
These structural features are similar to those in the MLL breakpoint cluster region (BCR). A DNA breakage and repair model has been proposed to explain nonhomologous recombination between MLL and its partner genes, particularly AF9.
