Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells and splenocytes, ≤ 0.6 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Internal Testing:
NovaFluor Yellow 610 shows non-specific staining of B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD44 is a 100 kDa type 1 transmembrane glycoprotein widely expressed on human leukocytes, brain white matter, and some epithelial cells. It serves as a receptor for hyaluronic acid and mediates cell-cell interactions, adhesion, and migration. CD44 is involved in lymphocyte activation, recirculation, and homing. Expression can be upregulated in certain carcinomas and may be linked to metastatic potential. Alternative splicing generates diverse isoforms contributing to functional variability.
