Target Information
ZRANB3 is a DNA annealing helicase and endonuclease required to maintain genome stability at stalled or collapsed replication forks. It facilitates fork restart and limits inappropriate recombination during template switching events (PubMed:21078962, 22704558, 22705370, 22759634, 26884333).
Recruited by polyubiquitinated PCNA to stalled replication sites, ZRANB3 acts as a structure-specific endonuclease that cleaves replication fork D-loop intermediates, generating a 3'-OH group suitable for DNA polymerase extension.
Additionally, ZRANB3 catalyzes fork regression via annealing helicase activity to prevent replication fork disintegration and double-strand break formation (PubMed:22705370, 22704558). [UniProt]
