Cellular Localization

Nucleus, Chromosome, Centromere, Kinetochore.
Localizes to the nucleus in absence of DNA damage.
Following DNA damage, recruited to double-strand break (DSB) sites by recognizing histone H2A monoubiquitinated at Lys-15 (H2AK15Ub) and histone H4 dimethylated at Lys-20 (H4K20me2).
Associated with kinetochores during mitosis.

Background

  • May play a role in checkpoint signaling during mitosis and enhances TP53-mediated transcriptional activation.
  • Involved in the cellular response to DNA damage.
  • Post-translational modifications (PTM):
    • Asymmetrically dimethylated on Arg residues by PRMT1; methylation required for DNA binding.
    • Phosphorylated at basal level without DNA damage; hyper-phosphorylated in ATM/ATR-dependent manner upon DNA damage.
  • Structure: Contains 2 BRCT domains.
  • Subcellular location: Nuclear and cytoplasmic; recruited to DSB sites.
  • Interactions:
    • Interacts with IFI202A, TP53/p53, DCLRE1C, histone H2AFX (phosphorylated on Ser-139), and histone H4 dimethylated at Lys-20.
    • Displays low or no affinity for unmethylated or trimethylated histones.
