Cellular Localization Cytoplasm, Nucleus, Cell junction, Synapse. In cortical neurons, predominantly cytoplasmic and associated with Golgi apparatus and endosomal fraction. Increased neuronal activity increases phosphorylated form at synapses. Colocalizes with POU5F1 in nucleus.

Background

The protein encoded by this gene belongs to the MAP kinase family, which integrates multiple biochemical signals and regulates cellular processes such as proliferation, differentiation, transcription, and development. It is activated by various stimuli and mediates immediate-early gene expression through transcription factor targeting. Activation by TNF-alpha is required for TNF-alpha–induced apoptosis and is also involved in UV radiation–induced apoptosis related to cytochrome c–mediated pathways. Mouse studies suggest a key role in T cell proliferation, apoptosis, and differentiation. Several alternatively spliced variants exist.
