Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extending into the extracellular space. Its receptor, FasR, is a cell-surface type-I membrane protein and a member of the tumor necrosis factor (TNF) and nerve growth factor (NGF) receptor family.
As a member of the TNF-cytokine family, CD178 induces apoptosis when interacting with FasR. It may exist as membrane-bound (45 kD) or soluble (26 kD) forms, with the soluble form released upon cleavage by metalloproteinases.
Binding of CD178 to Fas leads to receptor oligomerization and triggers apoptotic cell death through interaction with other proteins. CD178 is predominantly expressed in activated T-lymphocytes and natural killer (NK) cells, and also in immune-privileged tissues such as testis and eye.
Expression has been reported in thymus, liver, ovary, lung, heart, and kidney. Induction of apoptosis through CD178 is mainly involved in anti-viral immune responses.
The Fas/CD178 system plays a role in various human diseases, including AIDS, hepatitis, cancer, atherosclerosis, and inflammatory conditions related to hypertension.
