Target Information
Isocitrate dehydrogenases such as IDH1 catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one utilizing NAD(+) and the other NADP(+). Five isocitrate dehydrogenases have been reported: three NAD(+)-dependent forms localized to the mitochondrial matrix, and two NADP(+)-dependent forms, one mitochondrial and one cytosolic. The NADP(+)-dependent isozyme is a homodimer. The IDH1 protein is the cytoplasmic and peroxisomal NADP(+)-dependent isocitrate dehydrogenase containing a PTS-1 peroxisomal targeting signal. Its presence in peroxisomes suggests roles in NADPH regeneration for intraperoxisomal reductions and in reactions consuming 2-oxoglutarate. The cytoplasmic enzyme plays a key role in NADPH production. This antibody is specific for IDH1 (mutant R132H).
