Target Information

The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes from yeast to mammals. It is required for induction of RNA polymerase II transcription by DNA-binding transcription factors.
MED28 (also known as Magicin) is one of the proteins in this complex and is expressed in many cell lines and tissues. Down-regulation of MED28 in NIH3T3 cells induces genes associated with smooth muscle cell differentiation, while overexpression represses SMC gene expression.
MED28 can form a ternary complex with Grb2 and Merlin (the neurofibromatosis 2 tumor suppressor protein), suggesting a role in Merlin’s tumor suppressive activity. It has also been identified as an HIV dependency factor (HDF), making it a potential drug target in HIV treatment.
At least two isoforms of MED28 are known to exist.
